Olapani is a new oral poly ADP ribose polymerase (PARP) inhibitor. PARP includes the most important members of three protein families: PARP1, PARP2 and parp3. PARP enzymes are involved in normal cell homeostasis, such as DNA transcription, cell cycle regulation and DNA repair. The results of olaparine in vitro showed that the single drug treatment or platinum based chemotherapy could inhibit the growth of the selected tumor cell line and the tumor growth of human cancer mice xenotransplantation model. The cytotoxicity and antitumor activity of BRCA deficient mouse tumor cell lines were increased after treatment with olaparine. In vitro studies also showed that the cytotoxicity induced by olapamil may be related to the inhibition of PARP enzyme activity and the increase of parp-dna complex formation, leading to the destruction of cell dynamic balance and cell death.
